Imatinib is the first of a new class of drugs that act by specifically\u00a0inhibiting<\/a>\u00a0a certainenzyme<\/a>\u00a0\u2013 a\u00a0receptor tyrosine kinase<\/a>\u00a0\u2013 that is characteristic of a particular cancer cell, rather than non-specifically inhibiting and killing all rapidly\u00a0dividing cells<\/a>. Imatinib was a model for other\u00a0targeted therapies<\/a>\u00a0that inhibited this class of enzymes.<\/p>\n

It is used in treating\u00a0chronic myelogenous leukemia<\/a>\u00a0(CML),\u00a0gastrointestinal stromal tumors<\/a>(GISTs) and some other diseases. By 2011, Gleevec has been FDA approved to treat ten different cancers.<\/p>\n

In CML, the tyrosine kinase enzyme\u00a0ABL<\/a>\u00a0in white blood cells is locked in its activated form. This causes the excessive proliferation and high white blood cell count which is characteristic of CML. Imatinib binds to the site of tyrosine kinase activity, and prevents its activity, causing tumor cell death (apoptosis<\/a>).<\/p>\n
