Triptolide<\/strong> is a diterpenoid<\/a> epoxide<\/a> found in the Thunder God Vine, Tripterygium wilfordii<\/a><\/em>. It has in vitro<\/a><\/em> and in vivo<\/a><\/em> activities against mouse models<\/a> of polycystic kidney disease<\/a>[1]<\/a><\/sup> and pancreatic cancer<\/a>, but its physical properties<\/a> limit its therapeutic potential.[2]<\/a><\/sup> Consequently, a synthetic prodrug<\/a>, minnelide<\/a>, is being studied clinically<\/a> instead.[2]<\/a><\/sup><\/p>\n A little-known plant with a truly bizarre name is now making headlines as a cancer killer, with the compound of the plant vanishing tumors in mice with pancreatic cancer. Known as the \u2018thunder god vine\u2019 or lei gong teng, the Chinese plant is actually integrated into Chinese medicine and has been used for ages in remedying a number of conditions including rheumatoid arthritis.<\/p>\n According to the new research out of the University of Minnesota\u2019s Masonic Cancer Center, the thunder god plant compound led to no signs of tumors after a 40 day period \u2014 even after discontinuing the treatment. Its molecular mechanism of action, however, has remained largely elusive to date. We report that triptolide covalently binds to human XPB (also known as ERCC3), a subunit of the transcription factor TFIIH, and inhibits its DNA-dependent ATPase activity, which leads to the inhibition of RNA polymerase II\u2013mediated transcription and likely nucleotide excision repair. The identification of XPB as the target of triptolide accounts for the majority of the known biological activities of triptolide.
